I'm going to be sending out massive quantities of resumes to every single ad agency in the Philadelphia area.  Most of them don't have job postings right now and I won't be applying for a specific position, just expressing my interest in working there. What the heck do I say in a cover letter like this? Should I request an informational interview or something? Help, please! Thanks.

I would briefly highlight your experiences, and then detail your interest in working there...sort of explain why you're a good fit...try to make them remember you as being perfect for their company, so when they ARE hiring, they think of you. I wouldn't request anything.
